The comprehension of mechanisms regulating bacterial nucleoid structure and defining transcriptional cascade is still at the beginning, but could have strong epidemiological relevance. Indeed we hypothesize that nucleoid conformation regulation is the ground for all the processes involved in the regulation of the transition from vital bacteria to dormant coccoids, resistant to antibiotics but resurrectable in defined conditions. For this reason, we purpose to characterize nucleoid conformation in H.pylori, a class I carcinogen for gastric cancer, in several growth conditions including coccoid, with 4f-SAMMY-seq technology. This study will produce the first set-up of t4f-SAMMY-seq in bacteria, enabling the production of unbiased, high resolution, nucleoid accessibility maps in other species. Moreover it will provide the first characterization of H pylori nucleoid structure in several growth conditions paired with transcriptional data. This will be a fundamental step to start to dissect the mechanisms regulating nucleoid conformation.
